Trong "Jojos Bizarre Adventure: Stardust Crusaders", câu chuyện bắt đầu khi Jotaro Kujo, một học sinh trung học ngỗ ngược, bất ngờ phát hiện mình sở hữu "Stand" – một sức mạnh tâm linh siêu nhiên có hình dạng kỳ lạ. Cùng lúc đó, mẹ anh, Holly, cũng bị một Stand xuất hiện nhưng lại đe dọa đến tính mạng cô, tất cả là do sự trỗi dậy của ma cà rồng cổ đại, DIO, kẻ thù truyền kiếp của dòng họ Joestar.
Để cứu mẹ trước khi Stand hấp thụ hết sinh lực của cô, Jotaro cùng ông ngoại Joseph Joestar (người cũng là một người dùng Stand giàu kinh nghiệm) buộc phải bắt đầu một cuộc hành trình cam go. Chuyến phiêu lưu này sẽ đưa họ từ Nhật Bản băng qua nhiều quốc gia châu Á và Trung Đông, cuối cùng là đến Ai Cập – sào huyệt của DIO. Trên đường đi, họ kết nạp thêm những đồng minh mạnh mẽ như Kakyoin, Avdol, Polnareff và chú chó Iggy, đối mặt với vô số người dùng Stand độc ác được DIO phái đến cản đường. Mỗi trận chiến là một thử thách sống còn, đẩy Jotaro và nhóm bạn đến giới hạn để có thể đối đầu với DIO và chấm dứt mối hiểm họa này mãi mãi, giải cứu Holly.
